j.moreau, post #30: I read post #28 twice before replying, because I had assumed the opposite. Buffers and pH: the pH of a solution is buffered to maintain stability. The buffer system (phosphate, Tris, or other) is chosen to keep the solution at the pH where the peptide is most stable. Go to post

Isotonicity: an isotonic solution has the same osmotic pressure as blood. Non-isotonic solutions can cause discomfort on injection. Injectable peptide solutions are formulated to be isotonic.

I read post #31 twice before replying, because I had assumed the opposite.

Preservatives: solutions for multi-dose vials contain preservatives (benzyl alcohol, phenol, or others) to inhibit microbial growth after the vial has been entered. Single-use vials do not need preservatives.

Antioxidants: compounds that prevent oxidation (which damages peptides) are sometimes included. Methionine oxidation is a common degradation pathway and antioxidants help prevent it.

Two things before anyone answers the substance.

First, the context in the first post is clear and specific. Second, the question is framed so that an answer can actually address it. Both are the norm here and both matter more than they sound.

Amino acid residues and formulation: the specific sequence and composition of a peptide affects what formulation conditions it tolerates. Sequences high in methionine need more antioxidant protection. Sequences prone to aggregation need different pH or additive protection.

Coming back to post #35, because the follow-up matters more than the original answer.

Container closure: the vial material, rubber closure, and sealing all affect permeability to water and oxygen. Glass is less permeable than plastic. Rubber quality matters.

Fill volume and headspace: the amount of solution in a vial and the air above it affect how much the solution is exposed to oxygen during storage. Less headspace means less oxygen exposure.
